Identify The Focus
In นี่แหละคือสิ่งที่ผมหามานาน, the speaker identifies this as exactly what they have been looking for. คือ connects the identification to สิ่งที่ผมหามานาน, the thing sought for a long time.

People And Situations
คนนี้แหละที่เป็นแฟนฉัน identifies this person as the speaker's romantic partner. In ฉันนี่แหละที่จะไป, the emphasis falls on the speaker as the person who will go; whether anyone else can go depends on the situation. นี่แหละชีวิต comments on the situation as this is life or that's life.
